Characterization of extracellular vesicles derived from mesenchymal stromal cells by surface-enhanced Raman spectroscopy

Extracellular vesicles (EVs) are secreted by all cells into bodily fluids and play an important role in intercellular communication through the transfer of proteins RNA. There is evidence that EVs specifically released from mesenchymal stromal (MSCs) potent cell-free regenerative agents. However, for MSC to be used therapeutic practices, there must a standardized reproducible method their characterization. The detection characterization challenge due nanoscale size as well molecular heterogeneity. To address this challenge, we have fabricated gold nanohole arrays varying sizes shapes electron beam lithography. These platforms dual purpose trapping single enhancing vibrational signature surface-enhanced Raman spectroscopy (SERS). In paper, report SERS spectra derived pancreatic tissue (Panc-MSC) bone marrow (BM-MSC). Using principal component analysis (PCA), determined main compositional differences between these two groups found at 1236, 761, 1528 cm?1, corresponding amide III, tryptophan, in-plane -C=C- vibration, respectively. We additionally explored several machine learning approaches distinguish BM- Panc-MSC achieved 89 % accuracy, sensitivity, 88 specificity using logistic regression.
